He started his political career by working in politics and Dravidian political circles and community activities in the eastern districts of the city. The Dravida Munnetra Kazhagam party system helped him progress through its ranks, which enabled him to establish political connections throughout Chennai.

The beginning of his political career occurred when he won the Radhakrishnan Nagar constituency elections as a Member of the Tamil Nadu Legislative Assembly during the 2001 elections, which he repeated in 2006. He began his political career through two initial terms, which provided himwith legislative experience and enabled him to represent his constituents throughout urban Chennai.

Sekar Babu joined the Dravida Munnetra Kazhagam party in January 2011, and he ran for office under its political platform. His political advancement reached an important stage when he won the Harbour Assembly seat during the 2016 state elections, which he contested under DMK leadership.

He maintained his Harbour seat during the 2021 elections while voters continued to support him as he built his legislative power. The DMK returned to government during this time while Sekar Babu remained an Assembly member, which showed he still held a vital position in Tamil Nadu politics.

P. K. Sekar Babu received his appointment as Minister for Hindu Religious and Charitable Endowments HR&CE after the 2021 Tamil Nadu Assembly elections. He has held this position since 2026 under Chief Minister M. K. Stalin. The HR&CE Department manages temple affairs through which it controls the administration of temple operations and heritage preservation and their associated institutional frameworks.

Sekar Babu manages his responsibilities as HR&CE minister by overseeing the development of temple infrastructure through his work on modern facility introduction and expansion of accommodation services at major temple sites, which protect cultural heritage and provide public access.

P. K. Sekar Babu holds two positions, which include serving as MLA from Harbour and working as a cabinet minister in the Tamil Nadu government for his executive governance duties while implementing policies in his designated government responsibilities.
