Nanjil Sampath's origin was Manalikarai, Kothanallur Panchayat, Kalkulam taluk in Kanyakumari district, Tamil Nadu. He pursued his school education in his native place and further continued his education by getting M.A. and M.Phil degrees. The word "Nanjil" is the indicative of his home place, Nanjil Nadu.
He made his entry in politics in 1993 when he joined the Marumalarchi Dravida Munnetra Kazhagam (MDMK) right after the party's establishment under Vaiko. While in the ranks of MDMK, Sampath was known for his skills as a communicator and organiser, and he also took on leadership roles in the party, including propaganda and deputy general secretary.
After spending a year in opposition in 2011, he joined the ruling AIADMK in 2012. Later in 2012, he was elected an AIADMK deputy propaganda secretary. He held the office until early 2016. After a short period as a lone independent supporter of other political groups, he came back to active party politics and took up various roles, such as election campaign staff, during which he was a great supporter.
After 2019 and in the following years, Sampath actively worked to get the DMK candidates elected in the different elections of Tamil Nadu. He became a member of Tamilaga Vettri Kazhagam in December 2025, a party headed by the actor Vijay.
